Inactivity in obese mice linked to a decreased motivation to move

 

Starting a regular program at the gym is a common New Year's resolution, but it's one that most people are unable to stick with for very long. Now a study done in mice is providing clues about one of the reasons why it may be hard for so many people to stick with an exercise program. The investigators found that in obese mice, physical inactivity results from altered dopamine receptors rather than excess body weight. The report appears in Cell Metabolism on December 29.
"We know that physical activity is linked to overall good health, but not much is known about why people or animals with obesity are less active," says the study's senior author Alexxai V. Kravitz, an investigator in the Diabetes, Endocrinology, and Obesity Branch at the National Institute of Diabetes and Digestive and Kidney Diseases -- part of the National Institutes of Health. "There's a common belief that obese animals don't move as much because carrying extra body weight is physically disabling. But our findings suggest that assumption doesn't explain the whole story."
Kravitz has a background in studying Parkinson's disease, and when he began conducting obesity research a few years ago, he was struck by similarities in behavior between obese mice and Parkinsonian mice. Based on that observation, he hypothesized that the reason the mice were inactive was due to dysfunction in their dopamine systems.
"Other studies have connected dopamine signaling defects to obesity, but most of them have looked at reward processing -- how animals feel when they eat different foods," Kravitz says. "We looked at something simpler: dopamine is critical for movement, and obesity is associated with a lack of movement. Can problems with dopamine signaling alone explain the inactivity?"
In the study, mice were fed either a standard or a high-fat diet for 18 weeks. Beginning in the second week, the mice on the unhealthy diet had higher body weight. By the fourth week, these mice spent less time moving and got around much more slowly when they did move. Surprisingly, the mice on high-fat diet moved less before they gained the majority of the weight, suggesting that the excess weight alone was not responsible for the reduced movements.
The investigators looked at six different components in the dopamine signaling pathway and found that the obese, inactive mice had deficits in the D2 dopamine receptor. "There are probably other factors involved as well, but the deficit in D2 is sufficient to explain the lack of activity," says Danielle Friend, first author and former NIDDK postdoctoral fellow.
The team also studied the connection between inactivity and weight gain, to determine if it was causative. By studying lean mice that were engineered to have the same defect in the D2 receptor, they found that those mice did not gain weight more readily on a high-fat diet, despite their lack of inactivity, suggesting that weight gain was compounded once the mice start moving less.
"In many cases, willpower is invoked as a way to modify behavior," Kravitz says. "But if we don't understand the underlying physical basis for that behavior, it's difficult to say that willpower alone can solve it."
He adds that if we begin to decipher the physiological causes for why people with obesity are less active, it may also help reduce some of the stigma that they face. Future research will focus on how unhealthy eating affects dopamine signaling. The researchers also plan to look at how quickly the mice recover to normal activity levels once they begin eating a healthy diet and losing weight.

Parental obesity linked to delays in child development

Children of obese parents may be at risk for developmental delays, according to a study by researchers at the National Institutes of Health. The investigators found that children of obese mothers were more likely to fail tests of fine motor skill -- the ability to control movement of small muscles, such as those in the fingers and hands. Children of obese fathers were more likely to fail measures of social competence, and those born to extremely obese couples also were more likely to fail tests of problem solving ability.
The study, appearing in Pediatrics, was conducted by scientists at the NIH's Eunice Kennedy Shriver National Institute of Child Health and Human Development (NICHD).
"The previous U.S. studies in this area have focused on the mothers' pre- and post-pregnancy weight," said the study's first author, Edwina Yeung, Ph.D., an investigator in NICHD's Division of Intramural Population Health Research. "Our study is one of the few that also includes information about fathers, and our results suggest that dad's weight also has significant influence on child development."
Dr. Yeung and her coauthors cited research indicating that about 1 in 5 pregnant women in the United States is overweight or obese.
In the study, authors reviewed data collected from the Upstate KIDS study, which originally sought to determine if fertility treatments could affect child development from birth through age 3. More than 5,000 women enrolled in the study roughly 4 months after giving birth in New York State (excluding New York City) between 2008 and 2010. To assess development, parents completed the Ages and Stages Questionnaire after performing a series of activities with their children. The test isn't used to diagnose specific disabilities, but serves as a screen for potential problems, so that children can be referred for further testing.
Children in the study were tested at 4 months of age and retested 6 more times through age 3. When they enrolled, mothers also provided information on their health and weight -- before and after pregnancy -- and the weight of their partners.
Compared to children of normal weight mothers, children of obese mothers were nearly 70 percent more likely to have failed the test indicator on fine motor skill by age 3. Children of obese fathers were 75 percent more likely to fail the test's personal-social domain -- an indicator of how well they were able to relate to and interact with others by age 3. Children with two obese parents were nearly three times more likely to fail the test's problem solving section by age 3.
It is not known why parental obesity might increase children's risk for developmental delay. The authors note that animal studies indicate that obesity during pregnancy may promote inflammation, which could affect the fetal brain. Less information is available on the potential effects of paternal obesity on child development. The authors added that some studies have indicated that obesity could affect the expression of genes in sperm.
If the link between parental obesity and developmental delays is confirmed, the authors wrote, physicians may need to take parental weight into account when screening young children for delays and early interventional services.

People with obesity gene can still lose weight

 

People with a gene linked to weight gain are just as likely to benefit from weight loss programmes as those without, researchers have discovered.
The findings suggest diet, exercise and drug-based approaches to losing weight can be widely beneficial, even if some people may have a greater risk of piling on the pounds due to their genetics. In short, your DNA is not a barrier to weight loss.
While many genes are believed to affect body weight, a particular version of the so-called FTO gene shows one of the strongest associations with fat gain. Those carrying two copies of the genetic variant – about 16% of the population – are on average 3kg heavier than those without, and 1.7 times more likely to be obese.
While it is not known exactly how the genetic variant promotes weight gain, it is believed to increase the appeal of high-calorie foods and reduce the feeling of fullness after a meal. But whether it also affects efforts to shed pounds has been a matter of debate.
“It has become clear that genetics play a part in the reason why some of us get fatter,” said John Mathers, lead author of the research from Newcastle University. “The one that has the biggest effect in most people is the FTO gene, so we wondered whether having the [high-risk version of the] FTO gene would affect how well you could lose weight.”
Writing in the British Medical Journal, Mathers and an international team of collaborators describe how they analysed eight previously published randomised control trials involving a total of more than 9,500 overweight or obese adults to investigate whether carrying the obesity-linked version of the FTO gene affects the ability to lose weight.
In all of the studies, the participants were tested to discover whether they carried the genetic variant, and whether they had one copy or two, but the results were not disclosed to the participants. The researchers found that for each copy of the high-risk gene the participants possessed, they were, on average, almost 0.9kg heavier.
The participants took part in a variety of weight-loss programmes, including diet-based, exercise-based and drug-based approaches. “To our surprise, we discovered that carrying the [high-risk] FTO gene made no difference to your ability to lose weight. So people lost weight at just the same rate if they had the [high-risk version of the] FTO gene as if they didn’t,” said Mathers.
“There was no link between the type of the intervention – so whether the people were losing weight through diet or physical activity – and the gene. It seemed to work equally well.”
Sex and ethnicity, the authors said, did not affect the rate of weight loss, although they noted there was a lack of participants of Asian descent.
The studies, added Mathers, did not show whether carrying the obesity-linked version of the FTO gene affects whether weight loss was sustained, as the longest follow-up time was three years.
Dr Jude Oben, co-founder of the Obesity Action Campaign and senior lecturer in hepatology at University College London, welcomed the results. “Obesity is costing the NHS £16bn a year. We at Obesity Action Campaign are alarmed by this. Obesity causes cancer, diabetes, heart disease and liver cirrhosis. It is the HIV of our age. It is killing millions of our patients,” he said.
“That this size of study and its robust statistical methodologies support common sense is great. It means that general weight loss strategies which must involve the psychological, nutritional, physical and policy changes should be developed.”
Andrew Hattersley, professor of molecular medicine at the University of Exeter and part of the team that discovered the influence of the FTO genetic variant on weight gain, said it was not surprising that those with the variant were as able to lose weight as those without.
“Part of [obesity] is environmental, part of it is genetic, part of the genetic component is the FTO gene,” he said. “This isn’t a group that is remarkably different genetically. It is a very minor change and it is only a minor part of their susceptibility.” 

A number of so-called obesity genes have recently been uncovered. Some researchers have associated these genes with an increased difficulty in losing weight; however, a new review of existing data, published in The BMJ, calls this conclusion into question.

Although a wealth of obesity-related genes have been described, how much difference do they make?
Obesity has increased sharply over recent decades. Globally, an estimated 2.1 billion adults are now overweight or obese.
In the United States, more than one third of adults are obese.
The estimated medical cost of obesity in the United States was $147 billion in 2008.
In the interest of stemming this tide, much research has been plowed into methods of reducing this burden.
As genetic profiling has grown in speed and accuracy, our knowledge of the influence of genes on our bodies has increased in line.
Recent research into the genetics of obesity has uncovered a range of gene loci associated with body mass index (BMI) and the distribution of body fat. In fact, no less than 97 loci have been shown to account for 2.7 percent of variation in BMI.
The obesity-associated FTO gene, MC4R, and TMEM18 have shown the greatest association. And, of those top three, the FTO gene has been found to explain the largest amount of genetic variance in obesity traits.
Individuals with two copies of the FTO minor allele weigh an average of 3 kilograms more and have a 1.7-fold increased chance of being obese, compared with those who have two copies of the lower risk allele.
